ROLE OF TOXICOKINETICS IN IMPROVING DRUG SAFETY
Abstract
Toxicokinetic (TK) (concerning the absorption, distribution, metabolism, and excretion of toxic substances in an organism) can contribute significantly to increased drug safety. To limit toxicity and to ensure therapeutic success, it is crucial to understand the toxicological profile of compounds early in the drug development process. In this review, the significance of toxicokinetic to the safety of drugs was detailed, highlighting its value in the estimation and control of toxicity during preclinical, clinical, and regulatory studies. Through analysis of mechanisms of drug toxicity, the relationship between pharmacokinetics and toxicokinetic, and the role genetic and environmental factors play in toxicity, this article demonstrates how toxicokinetic data provide insight into dose optimization, patient safety and personalized medicine. We also review progress in predictive modelling, regulation, and approaches for minimizing toxicity through drug design and formulation. We also summarize predictive modelling, regulation, and strategies toward minimizing toxicity via drug design and formulation. Integration of toxicokinetic parameters into the drug development pipeline is necessary to generate safer drugs for patients and reduce the occurrence of adverse reactions to drugs.
